Centrifugal Pump: How It Works

How can I build a chemostat with a centrifugal pump?
I am trying to make a simple homemade chemostat that removes waste from a culture flask via a centrifugal pump and then have new media replenish the culture flask. I have drilled three holes in the top of a sealed container - two connected to tubing for the inflow and outflow and one for airflow. I currently have the outflow working with the pump. Is there a way to have fresh media flow in without an additional pump?
